PHTHALOCYANINIC PHOTOSENSITISERS FOR PHOTODYNAMIC THERAPY.

SYNTHESYS AND PHOTOPHYSICS

SENSO

General Objectives:

1. Selection of phtalocyanine compounds having photosensing properties, establishment of chemical synthese methods and prepare the substances. 2. Achievement of photophysical experiments and measurements in order to obtain experimental data on:

(i) comparative characterisation, through photophysical properties and constants, of the studied compounds, and comparative characterisation of generation of active species upon irradiation

(ii) establishing of the main elements of the irradiating scheme for every of the studied compound

3. Testing of biological cytotoxicity of the compounds of concern.

General objective are accompanied by stage objectives, e.g.:

•  methods for chemical synthesis and preparation of the substances

•  methods for laboratory measurement of photophysical constants

•  laboratory set-ups for measuring photophysical constants

•  influence of native quenchers over the fluorescence of the studied compounds

•  influence of binding over the photophysical constants involved in PDT

•  cell culture method.

 

The novelty of the project is granted by following aspects:

1. New phthalocyanic compounds

2. Methodically photophysical study of the proposed compounds, including measurement of the key photophysical constants involved in PDT

3. Paralel study of cytotoxicity and active species measurement (by direct physical methods)

4. Proposal of a new concept – the phtalocyanine Systema of photosensitisers.
